The Convergent Synthesis of Poly(glycerol‐succinic acid) Dendritic Macromolecules

Abstract: The high-yield convergent synthesis of dendrons, dendrimers, and dendritic-linear hybrid macromolecules composed of succinic acid, glycerol, and poly(ethylene glycol) (PEG) is described. This convergent synthesis relies on two orthogonal protecting groups; namely, the benzylidene acetal (bzld) for the protection of the 1,3-hydroxyls of glycerol and the tert-butyldiphenylsilyl (TBDPS) ester for protection of the carboxylic acid of succinic acid. These novel polyester dendritic macromolecules are composed entire… Show more

“…G1-core, G2-core, and G3-core were prepared using known procedures, 31,34 as shown in Scheme 1. First, the 1,3-diol of glycerol was protected as 1,3-benzylideneglycerol 1 by treatment with p-TsOH and benzaldehyde.…”

“…A solution of DIC (82 mL, 0.5 mmol) in dry DCM (0.5 mL) in a syringe was added in intervals over a period of 30 min. After 1 h additional stirring on the ice bath, the reaction mixture was placed in the refrigerator for 16 h. The mixture was transferred to a separation funnel and acetone (20 mL), water (20 mL) and O-tert-Butyl-G1-lithium carboxylate dendron (15). In a threenecked 100 mL reaction flask O-tert-butyl-G1-methyl ester dendron 6 (0.69 g, 1.43 mmol) and LiI (1.54 g, 11.5 mmol) were dissolved in dry THF (25 mL).…”
