The cytoskeleton of digitonin-treated rat hepatocytes.

Abstract: Treatment of isolated rat hepatocytes with low concentrations of digitonin increases the permeability of the plasma membrane to cytosolic proteins without causing release of organelles such as mitochondria into the surrounding medium. Electron microscopy showed that treatment of the cells with increasing concentrations of digitonin results in a progressive loss in the continuity of the plasma membrane, while most other aspects of cellular morphology remain normal. Depletion of background staining material from… Show more

“…To prevent the potential reverse transport of dopamine and thus "trap" cytosolic dopamine inside the cell, cultures were continuously exposed to the DAT inhibitor, mazindol. Cytosolic dopamine was then extracted using a mild, selective permeabilization of the plasma membrane with the steroid glycoside, digitonin (Fiskum et al, 1980). Using this digitonin-based permeabilization technique we extracted the cytosolic dopamine fraction from rat postnatal VM neurons transduced with VMAT2 overexpressing or VMAT2 silencing lentiviral vectors.…”

“…Mitochondrial and cytoplasmic enzyme fractions were separated by a digitonin method as described in Fiskum et al (13). Individual embryos were incubated in a 500-nl drop of enzyme extraction buffer (12), supplemented with 0.001% digitonin, for 2 min.…”

“…Protein extracts were prepared by selective plasma membrane permeabilization with digitonin to allow for separation of cytoplasmic, mitochondrial, and nuclear+membrane components according to a published protocol (Fiskum et al, 1980). Brie¯y, cells were washed in isotonic sucrose buer (250 mM sucrose, 10 mM KCl, 1.5 mM MgCl 2 , 1 mM EDTA, 1 mM EGTA, 10 mM HEPES, pH 7.1, plus protease inhibitors 1 mg/ml leupeptin, 2 mg/ml aprotinin, and 0.5 mM PMSF) and subsequently incubated for 1 min at room temperature in 100 ml of the same buer to which 0.05% digitonin has been added, followed by 10 min of centrifugation at 48C.…”
