The cytosolic calcium transient modulates the action potential of rat ventricular myocytes.

Abstract: SUMMARY1. The modulation of the action potential by the cytosolic Ca2+ (Ca0') transient was studied in single isolated rat ventricular myocytes loaded with the acetoxymethyl ester form of the Ca2+-sensitive fluorescent dye Indo-1. Stimulation following rest and exposure to ryanodine were used to change the amount of Ca2+ released from the sarcoplasmic reticulum and thus the size of the CaF' transient. The CaF+ transient was measured as the change, upon stimulation, in the ratio of Indo-1 fluorescence at 410 nm… Show more

“…68,69 This happens apparently via the increased Na ϩ /Ca 2ϩ -exchanger inward current that is boosted by augmentation of the calcium transients. 65 In rat myocytes, the Na ϩ /Ca 2ϩ -exchanger current is proportional to [Ca 2ϩ ] i. 70 The present study demonstrates that stretch influences the APs of the rat atrial myocytes.…”

“…Since calcium transient modulates the AP 65 and vice versa, 66 the changes in [Ca 2ϩ ] i during stretch should change the shape of the APs of the myocytes. It is known that inotropic interventions, such as stretch, increase the late duration of the APs 67 in cardiac myocytes with a short plateau.…”
