The D5 Dopamine Receptor Mediates Large-Conductance, Calcium- and Voltage-Activated Potassium Channel Activation in Human Coronary Artery Smooth Muscle Cells

Abstract: In addition to being an important neurotransmitter, dopamine exerts paracrine effects on the renal, adrenal, and gastrointestinal regulation of sodium balance and blood pressure.

“…However, this should similarly affect D 5 Ϫ/Ϫ mice syngenic or congenic transplanted and wild-type mice transplanted with wild-type or D 5 Ϫ/Ϫ kidneys, making unlikely a significant underestimation of the effect of nonrenal D 5 Rs on BP regulation. In the cardiovascular system D 5 Rs are expressed in smooth muscle of the tunica media of pial, pulmonary, coronary, and mesenteric artery branches, [45][46][47][48] and in vivo administration or in vitro application of D1-like receptor agonists induces vasodilation in the cerebral, coronary, and mesenteric vascular beds, reduces vascular resistance, and causes hypotension. Dopamine and its analogs, acting via D1-like receptors, are coronary vasodilators in animals and humans.…”

“…Dopamine and its analogs, acting via D1-like receptors, are coronary vasodilators in animals and humans. The vasodilation of coronary arteries, mediated by D 5 Rs, is attributed, in part, to activation of hyperpolarizing vasorelaxant potassium channels via cAMP/protein kinase G. 46,49 It is possible that the lack of the D 5 R-induced vasodilation is compensated by other vasodilatory systems that may include increased D 1 R function, although in other organs like brain and kidney, D 1 R function has not been shown to compensate for the lack of D 5 R. 16,50,51 In our studies, D 1 R expression is not altered by the absence of the D 5 R in renal and nonrenal areas.…”

“…Inhibition of Na + /K + ATPase activity in vascular smooth muscle cells would increase vascular resistance, as has been reported in the rat tail [99]. Low concentrations of dopamine, however, decreases systemic vascular resistance, probably by other mechanisms [100102], e.g., opening of potassium channels [103] that is mediated by D 5 R but not D 1 R, at least in human coronary arteries [104]. …”

“…White et al reported that BK Ca channels stimulation by natriuretic peptides through cGMP is due to the phosphorylation and activation of protein phosphatase 2A [133]. On the other hand, Natarajan et al [134] suggested that in human coronary artery SMC, activation of receptors increasing cAMP mediates the stimulation of the BK Ca channels via PKG [134]. These results imply that there may be cell-and tissue-specific BK Ca channels that are activated by PKG, one type activated by phosphorylation and the second type activated by dephosphorylation.…”
