The DARPin Encyclopedia: from Basic Research towards Therapeutics

Abstract: The adaptive immune system in vertebrates comes in two flavors. Historically, adaptive immunity focused on the study of immunoglobulins (also referred to as Ig-based antibodies herein); more recently, jawless vertebrates were found to use an entirely different class of proteins for their adaptive immunesystem. Specifically, these organisms express variable lymphocyte receptors (so called VLR antibodies) that create diversity based on the rearrangement of leucine-rich repeats. Meanwhile, various, naturally occu… Show more

“…1a ). DARPins were selected since they are stable, small, and disulfide-free α-helical repeat proteins that contain a single rigid and central paratope and fold robustly in fusion proteins 14,15,29 . They can be selected to bind and inactivate protein targets of interest, and function in both intracellular and extracellular environments.…”

“…Importantly, the concave paratope is flanked by constant N- and C-terminal cap regions for fusion of the photoswitchable domains. Finally, many DARPins have been reported against a diverse array of targets, and several DARPin-based therapies have entered clinical trials 14,15,29 . Thus, we expected that the successful discovery of a photoswitchable DARPins design would enable optical control over a variety of endogenous proteins.…”
