Experimental asthma in guinea pigs, as defined in 1937 by Kallós and Pagel, and the pertinent literature are reviewed. The relation between guinea pig asthma and human asthma bronchiale is established. The underlying mechanism in both processes is the misdirected (aberrant) formation of anaphylactic or reaginic (IgE) antibody against innocuous antigens, leading at renewed exposure to the antigen to release of mast cell mediators (such as histamine, SRS-A, eosinotactic peptides, PAF), bronchoconstriction, vasodilation and eosinophilic inflammation at the sites of antigen-antibody reactions in the tissues of the respiratory organs.

Precision-cut lung slices (PCLS) allow comparison of the airway responses of different species under identical experimental conditions. The aim of this study was to establish and characterise PCLS from guinea pigs (GPs) and to compare them with human PCLS.GP PCLS were prepared according to previously published procedures with the exception that the agarose solution and the initial incubation medium contained isoproterenol to avoid post mortem airway contraction.The median effective concentrations (EC50, expressed as nM) for agonist-induced bronchoconstriction in GP and human PCLS, respectively, were: leukotriene D 4 (1.8, 5.0); thromboxane (16, 1.3); serotonin (69, unresponsive); histamine (217, 2,170); and methacholine (231, 234). Allergen-induced bronchoconstriction of passively sensitised PCLS was attenuated by histamine or thromboxane-prostanoid receptor antagonists and was almost completely prevented by their combination with leukotriene receptor antagonists. Airways pre-contracted with methacholine were relaxed by the b-agonist salbutamol or the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine. Simultaneous studies of airways and vessels are possible with, for example, EC50 values for endothelin-1 of 37 nM (pulmonary arteries), 10 nM (pulmonary veins) and 9.6 nM (airway).When compared with previous findings in rat and mouse, these data show that guinea pig lungs are a more appropriate model for human airway pharmacology than lungs from rats or mice.

“…Though histamine can be released from intact mast cells, it is more likely that the increased vascular permeability mediated in monkey skin by human heat-stable IgG-type antibodies is analogous to the vascular changes induced in rabbits by antibody and antigen releasing histamine from platelets in the presence of complement (Henson & Cochrane, 1969a, b). In a previous experimental model of cot death, guinea-pigs were tested for their response to inhaled antigen (Parish, Barrett & Coombs, 1960), but death in these animals follows bronchoconstriction not observed in cot-death lungs (Parish, Akester & Gregg, 1964a). It is probable that rabbits which more readily form precipitins, and less readily form anaphylactic homocytotropic antibodies than guinea-pigs, would provide a more suitable experimental model.…”

Summary Sera of 50% of persons extremely sensitive and of 30% of persons moderately sensitive to milk had heat‐labile reaginic antibodies conferring passive cutaneous anaphylactic sensitivity on monkeys for 24 hr. More persons in each group had IgE antibodies that bound radio‐isotope labelled β‐lactoglobulin and passively sensitized monkey basophils for anaphylactic degranulation. These tests, however, are not certain evidence of clinical allergy, because of the sera of nineteen topic children, three mediated monkey PCA tests and five had γE anti‐$bt‐lactoglobulin antibodies detected by antigen binding or basophil degranulation, though the serum donors were not clinically allergic to milk. Similarly few normal, milk‐tolerant adults also had γE anti‐β‐lactoglobulin antibodies. Sera of nine children with milk‐induced gastroenteritis occurring 12–36 hr after exposure had no anti‐milk reagins, and only one had γE binding β‐lactoglobulin. This allergy may be mediated by heat‐stable IgG antibodies, because three sera conferred milk sensitivity on monkey skin for 4 hr and two others for 2 hr. The three sera had high titres of IgG agglutinins, one to β‐lactoglobulin, one to α‐lactalbumin and one to casein. All three fixed complement in the presence of milk, and one had precipitins. They did not passively sensitize human mast cells or monkey basophils for anaphylactic degranulation, but they prepared human lung for anaphylactic release of hist‐amine, though in small amounts compared to that released from lung prepared with reagins. The sera of two of three cases of cot death had IgG antibodies with similar properties.
