The dentate gyrus differentially metabolizes glucose and alternative fuels during rest and stimulation

Elisa M. York,
Anne Miller,
Sylwia A. Stopka
et al.

Abstract: The metabolic demands of neuronal activity are both temporally and spatially dynamic, and neurons are particularly sensitive to disruptions in fuel and oxygen supply. Glucose is considered an obligate fuel for supporting brain metabolism. Although alternative fuels are often available, the extent of their contribution to central carbon metabolism remains debated. Differential fuel metabolism likely depends on cell type, location, and activity state, complicating its study. While biosensors provide excellent sp… Show more

“…Samples of SOD1 A4V with or without 10-fold molar excess of S -XL6 in 10 mM Tris HCl (pH 7.4) were incubated for 4 h at 37°C [ 77 , 78 ]. After incubation, samples were alkylated with iodoacetamide (100 mM for 30 min), heated to 75°C for 20 min, and then treated with 2 volumes of Poroszyme immobilized trypsin (Applied Biosystems, Life Technologies Corporation, Carlsbad, California, USA) at 37°C for 15 min, mixing every few minutes to keep beads suspended.…”

“…Is it important? Well, it can happen, but in the grand scheme of things, no, it is not that important, as lactate is just as likely to efflux from neurons and is not a substrate that can maintain high level neuronal function ( Bak et al, 2006 , 2009 ; Kann, 2023 ; York et al, 2023 ). Giving it a name (ANLS) has given it an importance, which outstrips its role as one of many reasons that lactate enters or leaves cells and the brain.…”
