The diacylgylcerol-sensitive TRPC3/6/7 subfamily of cation channels: functional characterization and physiological relevance

Abstract: Among the "classical" or "canonical" transient receptor potential (TRPC) family, the TRPC3, -6, and -7 channels share 75% amino acid identity and are gated by exposure to diacylglycerol. TRPC3, TRPC6, and TRPC7 interact physically and coassemble to form functional tetrameric channels. This review focuses on the TRPC3/6/7 subfamily and describes their functional properties and regulation as homomers obtained from overexpression studies in cell lines. It also summarizes their heteromultimerization potential in v… Show more

“…Deletion of TRPC6 in mice resulted in the up-regulation of TRPC3 expression in vascular smooth muscle cells (44), which could interfere with the interpretation of our data. Human endothelial cells were shown to abundantly express TRPC6 mRNA and TRPC3 to a lesser extent (18).…”

“…TRPC channels, including TRPC1, TRPC3, TRPC5, TRPC6, and TRPC7, are expressed in vascular smooth muscle cells and may contribute to I cat (2,12,28,36,39,40,54). In cerebral artery smooth muscle cells, TRPC3 channel knockdown attenuated endothelin-1-and IP 3 -induced [Ca 2ϩ ] i elevation and constriction, indicating that this isoform is a major contributor to the IP 3 R-induced I cat in this cell type (54).…”

“…The closely related subgroup comprising TRPC3, TRPC6, and TRPC7 channels are directly activated by diacylglycerol through a PKC-independent mechanism (3,4). TRPC6 channels are highly expressed in a number of different tissues including vascular smooth muscle cells (5)(6)(7)(8). Despite their abundance, the exact physiological role of TRPC6 channels has not been elucidated.…”
