The different roles of cyclinD1-CDK4 in STP and mGluR-LTD during the postnatal development in mice hippocampus area CA1

Abstract: Background: Cell-cycle-related proteins, such as cyclins or cyclin-dependent kinases, may have functions beyond that of cell cycle regulation. The expression and translocation of cyclinD1-CDK4 in post-mitotic neurons indicate that they may have supplementary functions in differentiated neurons that might be associated with neuronal plasticity.

“…Importantly, expression levels and isoform expression of proteins with demonstrated roles in synaptic plasticity and Ca 2+ sensitivity have been shown to change during this maturational period, including, for example, adenylyl cyclases 1 and 8 (Conti et al. , 2007), CDK1/4 (Li et al. , 2007), and SNAP‐25 (Bark et al.…”

“…Activation of mGluR5 releases FMRP-mediated translation repression and results in protein synthesis-dependent LTD (Bear et al, 2004). Functional consequences of excessive mGluR signaling in absence of FMRP include prolongation of epileptic form bursts in hippocampal area CA3 (Bailey et al, 2008), elongation of dendritic spines on cultured hippocampal neurons (Vanderklish and Edelman, 2002), and LTD in hippocampal area CA1 (Li et al, 2007). These findings are associated with the FXS clinical phenotypes: epilepsy, elongated, and immature dendritic spines, and cognitive delay, according to the mGluR theory of FXS (Bear et al, 2004).…”

“…Several signaling molecules such as Rho (Welsh et al, 2001), JNK , NF-B (Guttridge et al, 1999) and the PI3K/AKT/mTOR/p70S6K1 pathway (Gao et al, 2003(Gao et al, , 2004 mediate the regulation of the expression and translocation of the cyclinD1-CDK4 complex in the hippocampus. It is postulated that mGluR-LTD activates the PI3K/AKT/mTOR cascade, which stimulates the cyclinD1-CDK4 complex via the retinoblastoma (Rb)/E2F1 pathway leading to modulation of protein synthesis (Li et al, 2007a) (Fig. 2).…”

“…Cyclins and cyclin-dependent kinases (CDKs) are not only involved in cell cycle control (Li et al, 2007a) but also have diverse roles in the regulation of basal transcription (Rickert et al, 1996), apoptosis (Crumrine et al, 1994), and synaptic plasticity (Richter, 2001). Several signaling molecules such as Rho (Welsh et al, 2001), JNK , NF-B (Guttridge et al, 1999) and the PI3K/AKT/mTOR/p70S6K1 pathway (Gao et al, 2003(Gao et al, , 2004 mediate the regulation of the expression and translocation of the cyclinD1-CDK4 complex in the hippocampus.…”
