The Differentiation and Maintenance of SARS-CoV-2-Specific Follicular Helper T Cells

Abstract: Upon acute viral infection, virus-specific CD4+ T cells differentiate into either TH1 cells or follicular helper T (TFH) cells. The molecular pathways governing such bimodal cell fate commitment remain elusive. Additionally, effector virus-specific TFH cells further differentiate into corresponding memory population, which confer long-term protection against re-infection of same viruses by providing immediate help to virus-specific memory B cells. Currently, the molecular mechanisms underlying the long-term ma… Show more

“…Additionally, ZC3HAV1 is an antiviral protein activated by interferons that can limit SARS-CoV-2 replication 27,28 . Furthermore, the involvement of CUL3 in regulating T follicular helper (TFH) cell differentiation 29 , was recently identified in the context of SARS-CoV-2 30 , suggests a mechanism for long-term protective immunity, indicative of vaccination success. While our study was not designed to elucidate the specific molecular mechanisms of their regulation, the fact that we consistently observe the regulation of these low-abundant immunity- and inflammation-related proteins is intriguing and suggests further exploration.…”

“…T cells, including helper T cell, cytotoxic T cells, and regulatory T cell, among which helper T cells can secrete a variety of cytokines, such as IFN-γ, IL-10, etc. to sensitize specific immune responses ( 40 ). In the present study, CSFV vaccine immunization significantly inhibited NK cell activity and promoted the secretion of IFN-γ and IL-10, while PTFP markedly enhanced NK cell activity and further facilitated IFN-γ and IL-10 secretion.…”

“…Strong T-cell responses are associated with milder COVID-19, and they also contribute to the protective response produced by vaccination ( 103 , 104 ). Studying SARS-CoV-2-specific T-cell responses could contribute to speeding up the vaccine testing process and facilitate vaccine development in the context of the COVID-19 pandemic ( 113 ). However, many unknowns remain, such as whether rapid induction of adaptive immune responses to SARS-CoV-2 and less severe COVID-19 manifestations are driven by pre-existing T cells ( 114 ), and whether a role for vaccine-elicited memory CD4 + T cell and CD8 + T cell responses in directly mediating protection from SARS-CoV-2 infection is unclear ( 115 ).…”
