The DNA-Damage Response to γ-Radiation Is Affected by miR-27a in A549 Cells

Abstract: Perturbations during the cell DNA-Damage Response (DDR) can originate from alteration in the functionality of the microRNA-mediated gene regulation, being microRNAs (miRNAs), small non-coding RNAs that act as post-transcriptional regulators of gene expression. The oncogenic miR-27a is over-expressed in several tumors and, in the present study, we investigated its interaction with ATM, the gene coding for the main kinase of DDR pathway. Experimental validation to confirm miR-27a as a direct regulator of ATM was… Show more

“…For example, ATM has been shown to be a gene target of a variety of miRNAs, including miR-223, miR-181a, miR26a, miR-27a, miR-214, and miR-18a [61][62][63][64][65][66][67]. These miRNAs enhance sensitivity to IR and etoposide by regulating the ATMmediated signaling pathway.…”

“…In addition, aberrant expression of miRNAs has been implicated in various aspects of cancer biology by regulating cancer cell survival, apoptosis, invasion and metastasis . With regard to the response of cancer cells to ionizing radiation (IR), an increasing number of IR‐responsive miRNAs and miRNAs that modulate cancer cell responses to IR have been identified .…”

“…for reviews of the relationship between miRNA expression and radioresponse. These changes often occur through alterations in: the expression of key DDR proteins; cell cycle regulatory proteins; regulation of cellular radical oxygen species; and proliferation of apoptosis genes such as p53, ATM, ATR, NFKB, and BCL2.…”
