The Dorsal Medial Habenula Minimally Impacts Circadian Regulation of Locomotor Activity and Sleep

Abstract: In nocturnal rodents, voluntary wheel-running activity (WRA) represents a self-reinforcing behavior. We have previously demonstrated that WRA is markedly reduced in mice with a region-specific deletion of the transcription factor Pou4f1 (Brn3a), which leads to an ablation of the dorsal medial habenula (dMHb). The decrease in WRA in these dMHb-lesioned (dMHbCKO) mice suggests that the dMHb constitutes a critical center for conveying reinforcement by exercise. However, WRA also represents a prominent output of t… Show more

“…Manual scores were assigned to 10 s epochs, for 3 h during the final third of the light phase; a time window with the highest probability to detect both stages of sleep, along with wake states. Next, we processed recordings during this same time interval using an autoscoring algorithm that was written in R (version 3.3.0) and described previously [87]. Manual scores were used as a standard to validate the autoscoring algorithm.…”

“…Sleep was recorded as previously described [35]. Briefly, mice were anesthetized with isoflurane and placed into a stereotaxic device where isoflurane anesthesia continued throughout surgery.…”

“…Raw ECoG/EMG data were automatically classified in 10-second bouts as either wake, NREM sleep or REM sleep using a previously described custom algorithm implemented in Python v2.7 [35]. Briefly, the power spectrums of the ECoG and EMG signals are calculated using Welch's method with an overlap window of 50%.…”

“…15 The DS mouse has been previously described to have deficits in both the circadian and homeostatic regulation of sleep. [15][16][17] Electrocorticographic Recordings Sleep was recorded as previously described 18 . Briefly, mice were anesthetized with isoflurane and placed into a stereotaxic device where isoflurane anesthesia continued throughout surgery.…”
