The Ecto-enzyme CD38 Is a Nicotinic Acid Adenine Dinucleotide Phosphate (NAADP) Synthase That Couples Receptor Activation to Ca2+ Mobilization from Lysosomes in Pancreatic Acinar Cells

Abstract: Nicotinic acid adenine dinucleotide phosphate (NAADP) is the most potent Ca 2؉-mobilizing intracellular messenger and is linked to a variety of stimuli and cell surface receptors. However, the enzyme responsible for endogenous NAADP synthesis in vivo is unknown, and it has been proposed that another enzyme differing from ADP-ribosyl cyclase family members may exist. The ectoenzyme CD38, involved in many functions as diverse as cell proliferation and social behavior, represents an important alternative. In panc… Show more

“…In addition, lysosomal disruption with bafilomycin A1 markedly reduced the fluorescence intensity of both GPR55 and Rab7, suggesting that GPR55 localizes intracellularly at the endolysosomes. GPR55 localization at this site correlates well with its ability to trigger NAADP-dependent Ca 2ϩ responses because NAADP-generating enzymes (70,71) have been also reported to localize at the membrane of acidic organelles (72).…”

“…Cells from the knockout mice no longer could elevate NAADP levels in response to IL8 [110]. Likewise, wild type pancreatic acinar cells responded to physiological concentrations of CCK with elevation of intracellular NAADP and Ca 2+ , but both were blocked in the acinar cells from the knockout mice [111].…”

“…However, the situation is more complex because in some cell types from CD38 knock-out mice, e.g. pancreatic acinar cells, NAADP levels are indeed lower compared with wild-type mice (16). Interestingly, increased NAADP contents in immunologically relevant tissues from CD38 knock-out mice, e.g.…”
