The Effect of Alkaline Earth Cations on the Release of Histamine From Rat Peritoneal Mast Cells Treated With Compound 48/80 and Peptide 401

Abstract: 1 Extracellular calcium ions have a dual effect on the release of histamine from rat peritoneal mast cells treated with compound 48/80 and peptide 401. The release is either potentiated or inhibited according to the relative concentrations of ion and inducer. 2 Strontium similarly potentiates the release produced by optimal concentrations of inducer but higher concentrations are required than in the case of calcium. Strontium is markedly less inhibitory than calcium. 3 Mast cells may be depleted of intracellul… Show more

“…Replacement of sodium with an equimolar concentration of lithium almost abolished the uptake and there was a tendency to an increased release of histamine as previously reported (Cochrane & Douglas, 1976;Atkinson et al, 1979). In presence of sodium the non-stimulated secretion was 5.1% + 1.9%, and in presence of lithium it was 8.2% + 3.8% (mean values and s.e.mean from five experiments, P < 0.1).…”

“…Mixed peritoneal cells were recovered by direct lavage (Atkinson et al, 1979) with modified Tyrode solution of the following composition (mM): NaCl 137, glucose 5.6, KCl 2.7, NaH2PO4 0.4, CaCl2 1 and N-2-hydroxyethyl piperazine-N'-2 ethane sulphonic acid (HEPES) 10. The pH of the solution was adjusted to 7.2 before use.…”

“…Individual supernatants were treated similarly. Histamine was then determined spectrofluorimetrically (Atkinson et al, 1979) using a commercial automated apparatus (Technicon Autoanalyzer II). Histamine release was expressed as a percentage of the total cellular content of the amine and was calculated as the ratio (histamine in supernatant)/(histamine in supernatant+ residual histamine in cells) (x 100).…”

“…Peritoneal cells were prepared by a method adapted from Atkinson et al (1979). Animals were killed by exposure to CO2, (100%) and Krebs Henseleit solution (KHS), 10 ml, was injected into the peritoneal cavity.…”
