The effect of different oils on the absorption of probucol in the rat

Abstract: The effect of oily vehicles on the gastrointestinal absorption of the hypocholesterolaemic agent probucol has been investigated in the rat. The plasma concentration was determined following its administration in arachis oil (peanut oil), Miglyol 812 (fractionated coconut oil) and liquid paraffin. The total absorption of the drug, calculated as the area under the plasma concentration time curve, was significantly greater for the arachis oil formulation than with the other vehicles. The drug was selectively abso… Show more

“…It was reported that probucol had high lymphatic bioavailability when coadministrated with lipids. 19,20 Chylomicrons (CMs) are important in drug delivery since many lipophilic drugs are carried by CMs and transported in lymph. 21 Palin and Wilson reported that digestion of oil within the small intestine resulted in the liberation of long-chain fatty acids, which effectively promoted chylomicron formation and consequently the lymphatic transport of DDT or probucol.…”

“…Oleic acid was chosen as the oil phase in all three lipid formulations as it is the principal long-chain fatty acid that occurs in human mixed intestinal lipids (Staggers et al 1990) and in recognition of the potential merits of longchain unsaturated fatty acids, particularly oleic acid, in the design of a lymphotropic lipid delivery systems (Palin & Wilson 1984;Charman & Stella 1986b).…”

“…Enhanced drug absorption was reported when using LCT (long chain triglycerides) compared to MCT (medium chain triglycerides) in SMEDDS (22,23); however, this cannot be taken as a rule.…”
