The effect of HCG on pinocytosis within the canine inter‐sertoli cell tight junction. A preliminary report

Abstract: The role of tight junctions in permeability barriers is now well established. Other roles for this junction have been postulated but have not been demonstrated. In both adult and prepubertal dogs, stimulation with human chorionic gonadotropin (hCG) results in an increased number of pinocytotic vesicles on the membrane faces enclosed by tight junctions within the inter-Sertoli cell junctions of the testis. It is suggested that these membranous regions, protected from the normal flow of intercellular fluids, ma… Show more

“…12 sexually mature mongrel dogs obtained from local suppliers were used in this study. Freeze-fracture replicas were prepared from cryoprotected fixed tissue (8), lightly fixed tissue (1 h in 1% Karnovsky's fixative [29] and cryoprotected with 20% glycerol in 0.1 M cacodylate buffer, pH 7.2, for 4-8 h), and from unfixed tissue not cryoprotected with glycerol, l-ram cubes of tissue mounted on 3-mm cardboard disks were frozen in Freon 22 (Phillips Manufacturing Co., Chicago, Ill.) suspended in liquid N2. The tissues were fractured at -115~ in a Balzers BA 360M freeze etch unit (Balzers High Vacuum Corp., Santa Ana, Calif.) and coated with platinum-carbon.…”

“…This sub-membrane structure can be seen as a meshwork of densely packed proteins in detergent-extracted cells [3]. Small vesicular structures and pinocytosis have been seen to associate with the tight junction, suggesting that it may be a region of selective intercellular exchange [4,5]. The membrane domain of the tight junction is subdivided into discreet corralled sub-domains, which are physically demarcated by polymerized membrane components.…”

“…Till date, occludin together with claudins has been found in testis and epididymis of the some mammals (Hirase et al 1997;Saitou et al 1997;Furuse et al 1998;Moroi et al 1998;Gow et al 1999;Morita et al 1999a,b;Hellani et al 2000;Gye et al 2003a). Although the morphological feature of the BTB was described in canines (Connell 1978), the molecular architecture of Sertoli and epididymal epithelial TJs is not yet uncovered.…”
