University of Helsinki, HelsinkiThe effect of carrier-primed cells on an adoptive immune response was studied. Carrier-primed cells came from donors immunized with chicken globulin (CG), and hapten-primed cells from donors immunized with NIP-BSA (4-hydroxy-3-iodo-5-nitro-phenylacetic acid coupled to bovine serum albumin). Recipients were boosted with NIP-CGG.Recipients of hapten-primed cells produced only 1jlOOOth of the anti-NIP produced by hapten-primed cells plus carrier-primed cells. The increase in anti-NIP titer was mainly "IgC".Recipients of only carrier-primed cells had 100 times higher anti-NIP titers than recipients of unprimed cells. This anti-NIP was mainly "IgM".Carrier-primed cells taken 8 days after one injection of CG enhanced the anti-NIP response more than cells taken from donors given two injections of CG. The latter, however, produced more anti-CG than the former.

Anti-NP [(4-hydroxy-3-nitro-phenyl)acetyl)] antibodies from the primary response of BALB/c mice contain a major idiotype NP-a that is controlled by an allotype-linked gene VHNPa. A new strain distribution pattern for a VH marker was discovered: strains BALB/c, RF, DBA, RIII and SM were positive, and strains C57BL/6, SJL, CBA, A/J, CE, AKR and NZB were negative for the idiotype NP-a. NP-a idiotype chared many characteristics with previously described idiotype NP-b and, therefore, allelism was suggested between them. Both idiotypes have lambda light chains, both seem to be conservative in evolution, both are predominant early in the antibody response. Later during the response, antibodies become increasingly idiotype-negative while at the same time, homogeneous isoelectric focusing patterns become very heterogeneous. The study of VH recombinant strains supported the concept of allelism because in every case studied, the idiotype were mutually exclusive. (C57BL/6 X BALB/c)F1 mice expressed idiotypes NP-a and NP-b regularly and in approximately equal amounts.

Idiotypic analysis of the response of C57BL/6 mice to the (4‐hydroxy‐3‐nitrophenyl)acetyl group

Strain C57BL/6 mice produce a highly restricted primary response to the (4-hydroxy-3-nitrophenyl)acetyl (NP) group. This response is composed of molecules having mu or gamma1 heavy chains and light chains of the lambda type. A guinea pig anti-idiotype was raised to the purified C57BL/6 primary anti-NP immunoglobulin. After suitable absorption, this anti-idiotype was shown to detect markers present on the primary anti-NP immunoglobulin only of those strains which express the Ig-1b allotype. Breeding experiments demonstrated that the marker segregated with the heavy chain linkage group.
