Starch is an inexpensive feed ingredient that has been widely used in fish feed. However, starch utilization by carnivorous fish is limited and excess starch is detrimental to the health of the organism. High starch diets often lead to liver damage, but the effects on the intestine are often overlooked. Therefore, in this study, two isonitrogenous and isolipidic semi-pure diets (NC: 0% α-starch, HC: 22% α-starch) were formulated and fed to largemouth bass (Micropterus salmoides) for 45 days. The effects of the high starch diet on the intestine of largemouth bass were comprehensively investigated by intestinal microbiota, histopathology, ultrastructural pathology, and enzymology analyses. Feeding the HC diet did not affect the growth of largemouth bass during the experimental period. However, the high starch diet led to a reduction in the diversity and abundance of intestinal microbiota in largemouth bass, with a significant increase in the abundance of harmful bacteria (Aeromonas) and a decrease in the abundance of beneficial bacteria (Clostridium, Lactobacillus, and Bifidobacterium). Feeding the HC diet caused the development of enteritis, with goblet cell hyperplasia, epithelial necrosis and detachment and inflammatory cell infiltration, and leading to enlarged apical openings and mitochondrial damage in goblet cells. Long-term feeding of the HC diet inhibited intestinal α-amylase activity. changes in the intestinal microbiota, such as an increase in Aeromonas and a decrease in Clostridium, Lactobacillus, and Bifidobacterium, may be closely related to the development of enteritis. Therefore, adding these beneficial bacteria as probiotics may be an effective way to prevent damage to the intestine of largemouth bass from a high carbohydrate diet. Our results suggest reducing the amount of starch added to the largemouth bass diets. This study provides a reference for protecting the largemouth bass gut during modern intensive culture.

Stocking density in Colossoma macropomum larviculture, a freshwater fish, in recirculating aquaculture system

This study aimed to investigate stocking density during larviculture of Colossoma macropomum in recirculating aquaculture system with slightly saline water. Densities of 10 (D10), 30 (D30) and 50 (D50) larvae/L were tested with five replicates each. Larvae were fed with Artemia nauplii during the first 15 days and with commercial diet from day 16 to day 30. Water was maintained at a salinity of 2.01 ± 0.41 g of salt/L, and performance and survival were evaluated throughout the experiment. At the end of the experiment, larvae of each tank were classified according to size as small—S (<18.0 mm and <0.08 g), medium—M (>18.0 mm and <22.2 mm, >0.08 g and <0.15 g) and large—L (>22.2 mm and >0.15 g), and the size distribution was calculated for each stocking densities evaluated. Larval weight, length and specific growth rate were not affected by the tested stocking densities throughout the experiment (p > .05). Heterogeneity was recorded at 30 days of larviculture for all tested densities with a predominance of class M animals. Survival during feeding with Artemia was not affected by the tested densities (p > .05). Survival during feeding with commercial diet was highest for D10 and lowest for D30 (p < .05). Global survival, considering the experiment as a whole, was similar among the tested stocking densities (p > .05). Larviculture of C. macropomum can be successfully carried out at stocking densities of up to 50 larvae/L during the first 30 days of rearing in slightly saline water in recirculating aquaculture system.

The rapid decline of longfin smelt Spirinchus thaleichthys, a threatened euryhaline forage fish in California, is a serious concern for scientists and resource managers. To recover and conserve this species, a captive culture program was initiated, focusing on the collection, captive rearing and breeding of wild broodstock, and the rearing of their offspring. Although progress has been made in the collection of broodstock and the production and culturing of larvae, no studies have evaluated the rearing of juvenile life stages in captivity. The present study examines methodological considerations for culturing F1 juvenile longfin smelt, specifically, the first efforts toward weaning juveniles to a dry commercial pellet feed. Cultured juvenile longfin smelt were fed live Artemia only or co-fed Artemia and dry feed for 62 days, and the effects of feed type on juvenile survival, growth, body condition, and fatty acid profiles were examined. No significant differences were observed between feeding treatments, despite an 80% reduction in Artemia in the co-feeding treatment. Furthermore, examination of fish stomach contents at the end of the trial confirmed the transition to dry feed. This is the first study to indicate successful feeding by longfin smelt on dry commercial pellets, and suggests that juvenile longfin smelt can be fully weaned onto dry feeds. Results of this study are critical for closing the lifecycle of longfin smelt in captivity and developing a successful conservation culture program for this imperiled species.
