Effect of Niceritrol on Streptozocin-Induced Diabetic Neuropathy in Rats

Niceritrol, a drug with peripheral tissue vasodilatory and serum lipid-lowering activity, was administered for 2 mo to rats with streptozocin-induced diabetes. Physiological and biochemical studies were subsequently conducted on rat nerve tissue. A markedly lower value of approximately 47% in sciatic nerve blood flow (SNBF) was detected in an untreated diabetic (DC) group than in a nondiabetic control group (CC). A significant delay in caudal motor nerve conduction velocity (MNCV) and significantly higher glucose, sorbitol, and fructose values were observed in the sciatic nerve and serum lipids. In contrast, a niceritrol-treated diabetic (DN) group had significantly higher SNBF, MNCV, and sciatic nerve myo-inositol values and lower serum triglyceride levels than group DC. No differences between these two groups were noted in glucose, sorbitol, and fructose levels in the sciatic nerve, or in cholesterol and glucose in serum. These findings suggest that niceritrol has a clear inhibitory effect on the development of delayed MNCV in the diabetic rat, which may be due to reduced nerve blood flow and/or decreased nerve myo-inositol levels.

Abstract-Regression mechanism of Ethyl 2-(p-chlorophenoxy) isobutyratc (C11113) in the arterial wall was studied for elucidation in terms of fatty acid-compositional change in cholesterol-fed rabbits. The following evidence was obtained: (1) The amount of cholesterol and cholesteryl ester in the atheromatous aorta were reduced after CPIB treatment.(2) CPIB resulted in a significant increase in the ratio of linoleate to oleate in both the arterial wall and serum after withdrawal of the cholesterol diet. It is suggested that there is a preferential hydrolysis of linoleate-rich cholesteryl ester in both the arterial wall and the serum. The purpose of this study was an attempt to elucidate the mechanism of reduction of cholesterol in atheroma in terms of FA compositional improvement after CPIB treatment.This is the first paper to describe an increase in ratio of linoleate to oleate in cholesterol fed rabbits treated with CPIB.

Some pharmacokinetic properties of nicotinic acid (NA) and niceritrol (pentaerythritoltetranicotinate) were investigated in rabbits. Niceritrol released NA at a faster rate on incubation in vitro with plasma or liver homogenate than in a pure buffer solution, indicating that niceritrol was actively hydrolyzed. After administration of niceritrol to rabbits no niceritrol was present in the plasma but metabolites, probably lower esters of niceritrol, free NA and nicotinamide were found. On oral administration of NA in a dose of 16 mg/kg higher maximal plasma levels of free NA (~6 μg/ml) were noted than after adminstration of niceritrol (1.4 μg/ml) in the same dose. A diet producing hyperlipaemia and containing 0.5% niceritrol elevated the levels of free NA in the plasma by about 2 μg/ml. It also reduced the increase in plasma cholesterol. The investigation suggests that the pharmacological and pharmacokinetic properties of niceritrol in the rabbit are similar to those in man.
