The effect of pentoxifylline on the flow properties of human blood

Abstract: Recent investigations have revealed that erythrocytes from patients with chronic arterial occlusive disease are significantly less deformable than red blood cells from healthy subjects. The influence of pentoxifylline on red blood cell fluidity was measured by a standard filtration technique using 8 micron membrane filters. Impaired deformability of erythrocytes was significantly improved in patients suffering from peripheral vascular disorders following intravenous injection of 200 mg pentoxifylline. “…On the other hand, elevation of the cAMP leads to cytosolic Ca 2+ decrease via inhibition of its entry into cells [43]. The Ca 2+ concentration increase was removed, if it was added to incubation medium preliminary phosphodiesterase inhibitor or a permeable cAMP analog [44][45][46]. It was found that all four PDE inhibitors -IBMX, vinpocetine, rolipram, pentoxifylline -decreased RBCA significantly and on the other hand they increased red cell deformability.…”

“…Marked alterations of the main haemorheologic parameters were recorded. On the basis of these findings we undertook this study with the following aims: 1) to investigate the rheologic properties of blood in a group of mountaineers after a HA expedition and 2) subordinately, to assess the possible effects on the same properties of a drug known to influence haemorheology (Pentoxifylline, Px, Trental~ 400) .4 4 Team, Expedition and Treatment We have investigated 14 mountaineers, members of the Italian expedition &dquo; To Annapurna II&dquo; (Himalaya of Nepal, 1981), immediately before their departure and after their return 61 days later. All subjects had given their informed consent to the study.…”

“…It is currently used in peripheral vascular disease. Although there is great enthusiasm for its potential use in acute stroke, no randomized trials have yet been done [9,[91][92][93].…”
