The effect of puerarin on serum nitric oxide concentration and myocardial eNOS expression in rats with myocardial infarction

Abstract: Two new lanostane triterpenoids, 29-hydroxypolyporenic acid C (4) and 25-hydroxypachymic acid (5), together with three known compounds, ergosta-7,22-dien-3beta-ol (1), polyporenic acid C (2) and pachymic acid (3), were isolated from the 95% ethanolic extract of the sclerotium of Poria cocos (Schw.) Wolf. Their structures were determined by extensive spectroscopic analyses, including IR, UV, ESITOF-MS, HRESI-MS, 1D and 2D NMR data (1H NMR, 13C NMR, 1H-1H COSY, NOESY, HSQC and HMBC).

“…VEGF and HIF are two of the important factors responsible for promoting angiogenesis, whereas increased eNOS activity stimulates the production of NO and triggers the endothelium-dependent vasodilatory cascade (Lambert et al, 2008). The enhanced eNOS activity and angiogenesis might be related to the augmentation of AKT/protein kinase B phosphorylation (Zhang et al, 2008b). A recent study revealed that AKT functions downstream of VEGF and promotes endothelial cells survival and mediated NO production via the direct phosphylation of eNOS (Hemmings, 2006).…”

“…Hence, regulation of intracellular ROS and modification of the apoptotic cascade may be an effective way to reduce pathological apoptosis. Flavonoids, a group of polyphenol compounds, constitute a major part of effective components in traditional Chinese herbal medicines, such as Ginkgo biloba, Panax notoginseng, and Radix Puerariae and so on, that are frequently used to treat cardiovascular disease [Mahady, 2002;Ng, 2006;Zhang et al, 2008;Mojzisova et al, 2009]. They demonstrate diverse biological and pharmacological effects, including anti-inflammatory, vasodilatation, and improving blood circulation, among others.…”

“…This traditional Chinese medicine has been used for the treatment of various diseases and to promote bone healing for millenia and recent data suggest beneficial activity (16)(17)(18)(19)(20). The active ingredients have been identified as the isoflavones: puerarin, daidzin and daidzein, which have estrogenic activity (21)(22)(23)(24).…”
