Vibrios belonging to the Harveyi clade are major pathogens of marine vertebrates and invertebrates, causing major losses in wild and cultured organisms. Despite their significant impact, the pathogenicity mechanisms of these bacteria are not yet completely understood. In this study, the impact of indole signalling on the virulence of Vibrio campbellii was investigated. Elevated indole levels significantly decreased motility, biofilm formation, exopolysaccharide production and virulence to crustacean hosts. Indole furthermore inhibited the three-channel quorum sensing system of V. campbellii, a regulatory mechanism that is required for full virulence of the pathogen. Further, indole signalling was found to interact with the stress sigma factor RpoS. Together with the observations that energy-consuming processes (motility and bioluminescence) are downregulated, and microarray-based transcriptomics demonstrating that indole decreases the expression of genes involved in energy and amino acid metabolism, the data suggest that indole is a starvation signal in V. campbellii. Finally, it was found that the auxins indole-3-acetic acid and indole-3-acetamide, which were produced by various (micro)algae sharing the aquatic environment with V. campbellii, have a similar effect as observed for indole. Auxins might, therefore, have a significant impact on the interactions between vibrios, (micro)algae and higher organisms, with major ecological and practical implications.

Aims: In this study, we investigated the effect of N‐acyl homoserine lactone‐degrading bacterial enrichment cultures (ECs) on larviculture of the giant freshwater prawn Macrobrachium rosenbergii. Methods and Results: The larval performance in terms of larval growth, larval survival, larval quality, duration of the larval rearing process and microflora levels in the rearing water as well as inside the prawn gut was investigated. The application of the EC bacteria was performed in two ways: by adding them directly into the larval rearing water and via enriched Artemia nauplii used for larval feeding. The results of the study demonstrated that both ECs that were tested had a similar positive effect on larval survival and larval quality, whereas they did not affect larval growth or the duration of the larval rearing process. Conclusions: Under normal hatchery conditions, the optimal EC densities were found to be 106 CFU ml−1 for adding into the rearing water and 5 × 108 CFU ml−1 for enrichment of Artemia nauplii used for feeding of the larvae. In the hatchery, the ECs can be grown on waste streams of Artemia hatching. Significance and Impact of the Study: Application of this kind of ECs could lead to a more sustainable aquaculture production, by replacing the use of antibiotics to control diseases.

Obtaining a better understanding of mechanisms involved in bacterial infections is of paramount importance for the development of novel agents to control disease caused by (antibiotic resistant) pathogens in aquaculture. In this study, we investigated the impact of catecholamine stress hormones on growth and virulence factor production of pathogenic vibrios (i.e. two Vibrio campbellii strains and two Vibrio anguillarum strains). Both norepinephrine and dopamine (at 100 μM) significantly induced growth in media containing serum. The compounds also increased swimming motility of the tested strains, whereas they had no effect on caseinase, chitinase, and hemolysin activities. Further, antagonists for eukaryotic catecholamine receptors were able to neutralize some of the effects of the catecholamines. Indeed, the dopaminergic receptor antagonist chlorpromazine neutralized the effect of dopamine, and the α-adrenergic receptor antagonists phentolamine and phenoxybenzamine neutralized the effect of norepinephrine, whereas the β-adrenergic receptor antagonist propranolol had limited to no effect. Finally, pretreatment of pathogenic V. campbellii with catecholamines significantly increased its virulence toward giant freshwater prawn larvae. However, the impact of catecholamine receptor antagonists on in vivo virulence was less clear-cut when compared to the in vitro experiments. In summary, our results show that—similar to enteric pathogens—catecholamines also increase the virulence of vibrios that are pathogenic to aquatic organisms by increasing motility and growth in media containing serum.
