The Effects of Anesthetics and Perioperative Medications on Immune Function: A Narrative Review

Abstract: Preclinical and clinical studies have sought to better understand the effect of anesthetic agents, both volatile and intravenous, and perioperative adjuvant medications on immune function. The immune system has evolved to incorporate both innate and adaptive components, which are delicately interwoven and essential for host defense from pathogens and malignancy. This review summarizes the complex and nuanced relationship that exists between each anesthetic agent or perioperative adjuvant medication studied and… Show more

“…In a study of the effects of anesthetics on NK cell activity and metastasis in a breast cancer mouse model, propofol was reported as the only agent that did not suppress NK cell activity or increase metastasis ( 100 ). Other intravenous agents such as thiopental, etomidate and ketamine, also have anti-inflammatory properties ( 101 ), and thiopental and ketamine were associated with suppressed NK cell activity ( 100 ).…”

“…However, even in malignancy without preceding inflammation, cancer-induced inflammation, secretions of inflammatory factors, and immune cell infiltration are main characters in tumor initiation and advanced metastasis [ 118 ]. Anesthesia and analgesia may impact cellular immunity and inflammation during surgery, and thus affect cancer outcomes [ 119 , 120 , 121 , 122 , 123 , 124 , 125 ].…”

“…The immune system consists of deeply connected and interrelated cellular networks and signaling for both non-specific and acquired cellular defenses (Fig. 1) [4]. Targeting cells, ligands, signaling This article is part of the Topical Collection on Anesthesiology and Critical Care molecules, and cytokines has become increasingly effective as cancer immunotherapeutics were applied to more and more patients [5].…”
