The effects of calcium antagonists on calcium overload contractures in embryonic chick myocytes induced by ouabain and veratrine

Abstract: 1 The protective effects of some calcium antagonists against different forms of calcium overload contracture were investigated in embryonic chick cardiac myocytes. 2 Tetrodotoxin-sensitive sodium currents were recorded from the myocytes by the whole-cell voltage-clamp technique. Although the peak current was attenuated by veratrine, the inactivation process was markedly inhibited, resulting in a large increase in the total inward current. Action potentials were prolonged by veratrine, automaticity was inhibite… Show more

“…HOE 694 is a new Na+-H+ exchange inhibitor which has potent cardioprotective and antiarrhythmic effects during ischemia and reperfusion (Scholz et al, 1993;Hendrikx et al, 1994). To further characterize the functional properties of HOE 694 on cardiac Na+ channels, we used veratrine intoxication of isolated cardiac tissues which is by now a well-established model of modified Na+ channel function-induced contracture (Patmore et al, 1989;Wermelskirchen et al, 1991). Effect of HOE 694 on action potential parameters in rat atria.…”

Therefore, veratrine intoxication is a model of late Na+ currentinduced diastolic contracture (Patmore et al, 1989;Le Grand et al, 1993). The major effect of veratridine is a sudden reduction of normal open-Na+ channel current.…”

“…Typical control responses were around 2-5 pm. The method used has been described in detail elsewhere (Patmore et al, 1989a (Patmore & Whiting, 1985;Patmore et al, 1989b). The effects of RS 30026 on edge movement are shown in Figure 3 together with the concentration-response relationship.…”

“…The salt solution contained (mM): NaCl 110, KCI 1, MgSO4 0.8, HEPES 19.3, NaHCO3 0.83 and dextrose 5.5, pH adjusted to 7.4 with NaOH. An area of the aggregate which contained an edge which displayed significant movement was selected for analysis, by use of a video motion detection system described previously (Patmore & Whiting, 1985;Patmore et al, 1989a (Marty & Neher, 1983 Nelder & Mead (1964), as implemented in the subroutine AMEOBA (Press et al, 1986).…”
