The effects of over-expression of the FK506-binding protein FKBP12.6 on K+ currents in adult rabbit ventricular myocytes

Abstract: This study examines the effects of the intracellular protein FKBP12.6 on action potential and associated K+ currents in isolated adult rabbit ventricular cardiomyocytes. FKBP12.6 was over-expressed by ~6 times using a recombinant adenovirus coding for human FKBP12.6. This over-expression caused prolongation of action potential duration (APD) by ~30%. The amplitude of the transient outward current (Ito) was unchanged, but rate of inactivation at potentials positive to +40 mV was increased. FKBP12.6 over-express… Show more

“…the number of virus particles per cell. Efficient infection of adult cardiomyocytes can generally be attained with an MOI of 100 [108110]. Similar MOI values can be employed in neonatal cells [108, 111], although efficient infection may also be possible at lower virus concentration (MOI = 10–20 in [112, 113]).…”

“…34 Overexpression of FKBP1A by adenovirus-transfected isolated cardiomyocytes increases the SR Ca 2+ content and decreases Ca 2+ spark frequency. 35 Calmodulin acts as part of a calcium signal transduction pathway, mediating the control of many enzymes, ion channels and other proteins through calcium binding. 36 Among the enzymes to be stimulated by the calmodulin-calcium complex are numerous protein kinases, such as myosin light-chain kinases.…”
