The Effects of Pterostilbene on Neutrophil Activity in Experimental Model of Arthritis

Abstract: It has been demonstrated that pterostilbene inhibits reactive oxygen species production in neutrophils in vitro. However, little is known about its effects on neutrophils during inflammation in vivo. In this study, the effect of pterostilbene on neutrophil activity was investigated in experimental arthritis model. Lewis rats were injected by a single intradermal injection of heat-killed Mycobacterium butyricum in Freund's adjuvant to develop arthritis. Another group of arthritic animals received pterostilbene … Show more

“…The anti-rheumatic activity of PTS had been attempted in rat adjuvant induced arthritic model (Bauerova, Ponist, Mihalova, Drafi, & Kuncirova, 2011;Macickova et al, 2010;Perecko et al, 2013). However, daily oral dose of PTS at 30 mg/ kg did not exhibit much disease modifying effect in adjuvantinduced arthritic rats (Bauerova et al, 2011;Macickova et al, 2010;Perecko et al, 2013).…”

“…However, daily oral dose of PTS at 30 mg/ kg did not exhibit much disease modifying effect in adjuvantinduced arthritic rats (Bauerova et al, 2011;Macickova et al, 2010;Perecko et al, 2013). PTS did not suppress oedema due to inflammation although it lowered the number of neutrophils in blood (Bauerova et al, 2011;Macickova et al, 2010;Perecko et al, 2013). The marginal anti-rheumatic effects could be attributable to formulation problems and/or insufficient dosing.…”

“…RES can also exert a protective effect against other types of toxicity such as ethanol and glutamate [37–39]. PTER is a compound structurally similar to RES that also exhibits comparable antioxidant, anti‐inflammatory, and anti‐carcinogenic properties [15,40,41]. However, PTER has increased lipophilic and oral absorption compared with RES due to the presence of two methoxy groups; 80% bioavailability for PTER, whereas only 20% bioavailability for RES [11,12]…”

“…PTER exhibits higher bioavailability because it is more easily transported into the cell and more resistant to degradation and elimination [12]. Most research conducted on PTER function thus far has been focused on its anti‐cancer and anti‐arthritis activities [13–15]. Recently, it was reported that PTER also exhibits neuroprotective effects [16].…”

“…Each experimental group consisted of 10 animals and the substances tested were applied over a period 21 days from arthritis initiation. Then the formation of reactive oxygen species (spontaneous and stimulated with PMA) was determined on the basis of luminolenhanced chemiluminescence [25,26]. The samples contained 50 μl aliquots of 1.25 mmol/l luminol, 40 U/ml horseradish peroxidase, rat blood diluted 200-times with Tyrode's solution, 0.05 μmol/l PMA (or PBS) and Tyrode's solution.…”
