The effects of serotonin on the electrophysiological properties of atrioventricular node during an experimental atrial fibrillation

Abstract: A few studies explored the atrioventricular (AV) nodal effects of 5-hydroxytyptamine (serotonin, 5-HT) during supraventricular tachyarrhythmia. The aims of the present study are to investigate (i) 5-HT effects on the rate-dependent electrophysiological functions of AV node during atrial fibrillation (AF) and (ii) the potential contribution of various 5-HT receptors and the role of the autonomic nervous system on 5-HT effects on AV nodal properties. The specific stimulation protocols were applied to detect the … Show more

“…Therefore, drugs targeting on the serotonin system, such as serotonin receptor agonist and antagonists, are used in the treatment of motility disorders. Although these drugs can relieve the patients' symptoms, there are also some dangerous adverse effects, such as arrhythmia [ 16 , 17 ]. Thus, it is crucial to find a new therapeutic approach to relieve gastrointestinal dysmotility associated with obesity.…”

“…Interestingly our patient had no structural heart disease but has a past history of migraine and takes rizatriptan, a serotonin (5-HT) receptor agonist. 5-HT has been shown to cause prolongation of the nodal refractoriness more than atrial-His conduction time which leads to an increase in the excitability index without significant reduction of the ventricular rates during AF [ 10 ]. The presence of comorbidities like coronary atherosclerosis, obesity, dyslipidemia, hypertension, and hypothyroidism in our patient may have led to a predisposition for increased sympathetic tone, autonomic dysregulation, and increased sensitivity of the cardiac tissue to adenosine leading to paroxysmal atrial fibrillation that was unmasked by IV adenosine administration during cardiac catheterization [ 5 ].…”

“…Increased cytosolic IP 3 causes sarcoplasmic reticulum (SR) Ca 2+ release via IP 3 ‐receptor ligand‐gated Ca 2+ channels, leading to tachycardia and arrhythmia in various vertebrate models (Bramich et al., ; Centurión et al., ; Villalón and Centurión, ; Zima and Blatter, ). Serotonin was also shown to induce β‐adrenergic receptor activity in the rabbit atrioventricular node (Changizi et al., ).…”
