The effects of zoledronic acid on ECG: a prospective study on patients with bone metastatic cancer

Abstract: SummaryIntroduction. There are controversial results in the risk of atrial fibrillation as well as arrhythmogenic potential of bisphosphonates. Method. 37 patients and 40 healthy controls were evaluated prospectively with regard to the cardiac side effects related to the use of zoledronic acid (ZA) and its effects on electrocardiography (ECG) parameters. Result. As the basal ECG results of the patients diagnosed with cancer compared with the control group, it was determined that QT maximum was significantly lo… Show more

“…One of the cardiovascular side effects of high‐dose ZOL treatment is the increased risk of atrial fibrillation events. The potential mechanism has been attributed to altered intracellular ion concentration and pro‐inflammatory, pro‐fibrotic, and anti‐angiogenic properties of BPs . In addition, according to the World Health Organization (WHO) causality scale, ZOL is a probable cause of acute severe diarrhea and subsequent hypokalemia .…”

“…Women comprised the majority of the cohorts, making up 87.5% of the total population of meta-analyzed data. Indications for bisphosphonates varied; four studies focused on patients with primary osteoporosis, (18)(19)(20)(21) one on new diagnoses of bone metastases, (22) and one on osteoporosis secondary to cerebral palsy in children. (23) All studies provided pre-and post-infusion outcome data.…”

“…(23) All studies provided pre-and post-infusion outcome data. Five of the studies used zoledronic acid, (18)(19)(20)(21)(22) whereas one used pamidronate. (23) Length of follow-up for ECG monitoring varied greatly between studies and ranged from 24 hours to 30 days post-bisphosphonate infusion.…”
