The Egr-1 transcription factor directly activates PTEN during irradiation-induced signalling

Abstract: The PTEN tumour suppressor and pro-apoptotic gene is frequently mutated in human cancers. We show that PTEN transcription is upregulated by Egr-1 after irradiation in wild-type, but not egr-1-/-, mice in vivo. We found that Egr-1 specifically binds to the PTEN 5' untranslated region, which contains a functional GCGGCGGCG Egr-1-binding site. Inducing Egr-1 by exposing cells to ultraviolet light upregulates expression of PTEN messenger RNA and protein, and leads to apoptosis. egr-1-/- cells, which cannot upregul… Show more

“…In contrast, p53 activates PTEN transcription by directly binding to a p53-binding element in the PTEN promoter (Stambolic et al, 2001). Egr1 has been reported to be a crucial PTEN transactivator by directly binding to the PTEN 5 0 -untranslated region (Virolle et al, 2001). In this study, we show that Egr1 provides a critical link between b-catenin/TCF signaling, induced by the loss of E-cadherin, and PTEN transcription.…”

“…Similarly, PTEN promoter activities were repressed by E-cadherin loss and could be restored by mouse E-cadherin overexpression in OVCAR-3 and SKOV-3 cells ( Figure 4D), suggesting that E-cadherin can regulate PTEN at the transcriptional level. As transcription of PTEN can be transactivated by Egr1, by binding to an Egr1-binding site in the PTEN promoter (Virolle et al, 2001), we examined the protein levels of Egr1 in transfected SKOV-3 and OVCAR-3 cells. Egr1 protein levels were reduced in E-cadherin-depleted cells, and could be restored by the overexpression of mouse E-cadherin (Figures 4B and C), suggesting that Egr1 may mediate the effects of E-cadherin on PTEN transcription.…”

“…The PTEN gene transcription is regulated by diverse transcriptional factors. C-Jun binds to the AP-1 site of the PTEN promoter and can suppress its transcription, whereas Egr-1 activates PTEN transcription by direct binding to the PTEN 5 0 untranslated region (Virolle et al, 2001). In addition, p53 has been reported as a crucial PTEN transactivator (Stambolic et al, 2001).…”
