The establishment of new protein expression system using N starvation inducible promoters in Chlorella

Abstract: Chlorella is a unicellular green microalga that has been used in fields such as bioenergy production and food supplementation. In this study, two promoters of N (nitrogen) deficiency-inducible Chlorella vulgaris N Deficiency Inducible (CvNDI) genes were isolated from Chlorella vulgaris UTEX 395. These promoters were used for the production of a recombinant protein, human granulocyte-colony stimulating factor (hG-cSf) in Chlorella vulgaris UTEX 395 and Chlorella sp. ArM0029B. To efficiently secrete the hG-cSf, … Show more

“…High-capacity gene stacking toolkits, e.g., Golden Gate Modular Cloning (MoClo), have also been established for Nannochloropsis and Chlamydomonas strains, thus, providing a library of molecular building blocks for the genetic engineering of microalgae cells [ 60 , 61 ]. The Characterization and design of microalgae-specific constitutive, inducible, and synthetic promoters have also improved the flexibility and efficiency of the bioengineering process [ 62 , 63 ]. These molecular tools can be employed to target any desired DNA sequence for the generation of knockout, knockdown, or insertion mutants.…”

“…Chlorella, a common single-celled green algae, has a fast growth and reproduction rate and is only organism on earth that can grow four times within 20 hours (Shi et al 2020;Miyazawa et al 2020).…”

“…Whether alanine contributed to TAG accumulation was examined using inhibitors targeting on alanine catabolism. The overexpression of a gene encoding the alanine catabolism key enzyme, alanine aminotransferase (ALT), driven by a Chlorella vulgaris N De ciency Inducible (CvNDI1) promoter 10 together with the pharmaceutical inhibition of ALT activity was allowed us to con rm the alanine catabolism to lipid biosynthesis. Because of higher growth and CO 2 xation rates, microalgae rich in carbohydrates (without lignin and hemicelluloses) make the best feedstock for the production of bioethanol 2,11 .…”
