The estrogenic endocrine disrupting chemical bisphenol A (BPA) and obesity

Abstract: There is increasing experimental and epidemiological evidence that fetal programming of genetic systems is a contributing factor in the recent increase in adult obesity and other components of metabolic syndrome. In particular, there is evidence that epigenetic changes associated with the use of manmade chemicals may interact with other factors that influence fetal and postnatal growth in contributing to the current obesity epidemic. The focus of this review is on the developmental effects of estrogenic endocr… Show more

“…The strong estrogens that supported MCF-7 cell proliferation at subnanomolar concentrations included 17β-estradiol (E2), diethylstilbestrol (DES) (17), and 17α-ethynylestradiol (EE2) (18). Estrogens with intermediate strength supported MCF-7 cell proliferation at 1-100 nM and included two representative industrial chemicals BPA and p-nonylphenol (PNP) (1,19). The isoflavone phytoestrogens genistein and daidzein, which require metabolic activation for their full-strength estrogenic actions (20), were weak estrogens that supported MCF-7 cell proliferation at 0.1-10 μM.…”

“…T he endocrine disrupting chemicals (EDCs) are environmental pollutants that interfere with the endocrine system to disturb biological processes such as development, reproduction, and metabolism (1)(2)(3)(4). EDCs consist of a wide variety of manmade and natural compounds with highly diversified chemical structures (4).…”

“…The etiology of obesity involves numerous factors that can be defined as hereditary, congenital, social and cultural (Kopelman and Grace, 2004;Ogden et al, 2006). There is growing experimental and epidemiological evidence that fetal programming of genetic systems is a contributing factor in the recent global increase in adult obesity (Vom Saal et al, 2012).…”
