The Examination of Sulphur Auxotrophs: A Warning

Abstract: SUMMARYThe chemical instability and reactivity of certain biologically important sulphur derivatives are recalled and their relevance to the study of sulphur auxotrophs is pointed out.In the early stages of the biological utilization of inorganic sulphur sources the compounds involved are often relatively unstable. In the conditions used for the examination of their specific biological effects the compounds themselves, even when initially pure, may undergo chemical change by reaction with other compounds in gr… Show more

“…Supplementing the minimal medium with thiosulfate but not with sulfate or sulfite allowed strain 53RA3 to grow. No growth was obtained on cysteine-S-sulfinic acid, a probable intracellular source of sulfite (17), or on metabisulfite, a more constant source of extracellular sulfite (24). Very slow growth was achieved on a medium containing sulfide.…”

“…Sulfur compounds were obtained from Sigma Chemical Co. (St. Louis, Mo.) or Aldrich Chemical Co., Inc. (Milwaukee, Wis.), and to minimize decomposition (24), they were dissolved in water just prior to use, filter sterilized, and added to cooled-down R2MM-S agar. Plasmids pKC505 (26) and pAL7180 (12) were obtained from Nagaraja Rao, Eli Lilly Laboratories, and from Leonard Katz, Abbott Laboratories, respectively.…”

“…Trudinger (19640), however, has poiuted out that a partial resolution of the reaction sequence could result duriug extraction of the bacteria and that reactions may be modified by the presence of excess reduced glutathione, which is required for thiosulphate oxidation by cell-free extracts. The necessity for considering the possible effects of non-specific chemical interactions between inorganic sulphur compounds and thiols has recently been emphasized by Postgate (1963). There are, moreover, some obvious metabolic differences between Thiobacillus X and the straiu of T. thioparus used by Peck.…”

“…denitrificans were tested on minimal agar supplemented with the appropriate sulphur source. Because of the instability of cysteine, sodium sulphite and sodium thiosulphate (Postgate, 1963), these solutions were always made up just before use and sterilized by filtration through Millipore filters (0.4 pm pore size).…”
