The gateway to guanine nucleotides: Allosteric regulation of IMP dehydrogenases

Abstract: Inosine 5′‐monophosphate dehydrogenase (IMPDH) is an evolutionarily conserved enzyme that mediates the first committed step in de novo guanine nucleotide biosynthetic pathway. It is an essential enzyme in purine nucleotide biosynthesis that modulates the metabolic flux at the branch point between adenine and guanine nucleotides. IMPDH plays key roles in cell homeostasis, proliferation, and the immune response, and is the cellular target of several drugs that are widely used for antiviral and immunosuppressive … Show more

“…We also found that human IMPDH1 is a member of this Class II and is capable of forming fibers in the presence of MgATP (Labesse et al, 2013). Ultimately, the characterization of other IMPDHs from eukaryotic organisms reported by other groups also showed identical regulation by MgATP as of Class II bacterial IMPDHs, that is, for IMPDH of fungi Ashbya gossypii (Buey et al, 2017), and human IMPDH1 and IMPDH2 isoforms (Anthony et al, 2017; Buey et al, 2022; Burrell et al, 2022).…”

“…We also identified two types of metabolic enzymes as SARs, Nitrilase and IMPDH. NIT hydrolyzes nitrile groups into acids via thiol-mediated catalysis 60 and IMPDH catalyzes the oxidative reaction of IMP to xanthosine 5′-monophosphate (XMP), the rate-limiting step in the synthesis of guanine nucleotides 61 . These enzymes are found in prokaryotes and eukaryotes and, although they seemingly have nothing in common, both belong to a growing list of filament-forming enzymes whose activity is affected by their oligomeric state 62,63 .…”

“…These nucleotides assume vital roles in numerous biological processes, encompassing energy metabolism, DNA and RNA synthesis, and cellular signaling, among other physiological functions ( Buey et al, 2015 ). Therefore, the regulation of the de novo and salvage pathways of guanine nucleotides is crucial for maintaining the balance of adenine and guanine nucleotides and energy status in the cell ( Hyle et al, 2003 ; Buey et al, 2022 ).…”
