The general amino acid control regulates MET4, which encodes a methionine‐pathway‐specific transcriptional activator of Saccharomyces cerevisiae

Abstract: A met4 mutant of Saccharomyces cerevisiae was unable to transcribe a number of genes encoding enzymes of the methionine biosynthetic pathway. The sequence of the cloned MET4 gene allowed the previously sequenced flanking LEU4 and POL1 genes to be linked to MET4 into a 10,327 bp contiguous region of chromosome XIV. From the sequence and mapping of the transcriptional start points, MET4 is predicted to encode a protein of 634 amino acids (as opposed to 666 amino acids published by others) with a leucine zipper d… Show more

“…This same protein has been also recently identi®ed as an interactor of the HIV-1 viral protein Vpu (Margottin et al, 1998). b-Trcp is the human homolog of Xenopus b-TrCp (94% overall identity at the amino acid level; 96% similarity) (Spevak et al, 1993), Drosophila Slimb (77% id, 87% si) (Jiang and Struhl, 1998), C. elegans LIN-23 (57% id, 69% si) (Kipreos et al, 1999), and probably of S. cerevisiñ Met30 (28% id, 45% si) (Mountain et al, 1993). The function of b-TrCp homologs is intimately connected with cell proliferation.…”

“…It is known that the transcription of the MET4 gene itself is not repressed by the presence of methionine, but is regulated by the general amino acid control mechanism, and that weak stimulation of the MET4 transcript level is observed when 2-21) mM methionine is added to the media [10]. Therefore, the elevation in the wild-type MET4 mRNA level in the FDM441 strain in the presence of 5 mM methionine (Fig.…”

“…Met4p belongs to the basic region-leucine zipper (bZIP) protein, and mediates transcriptional activation of sulfur genes [9,10]. Functional dissection of Met4p has identified domains required for transcriptional activation and susceptibility to negative regulation via AdoMet [11].…”
