The General Transcription Repressor TaDr1 Is Co-expressed With TaVrn1 and TaFT1 in Bread Wheat Under Drought

Abstract: The general transcription repressor, TaDr1 gene, was identified during screening of a wheat SNP database using the Amplifluor-like SNP marker KATU-W62. Together with two genes described earlier, TaDr1A and TaDr1B, they represent a set of three homeologous genes in the wheat genome. Under drought, the total expression profiles of all three genes varied between different bread wheat cultivars. Plants of four high-yielding cultivars exposed to drought showed a 2.0–2.4-fold increase in TaDr1 expression compared to… Show more

“…Amplifluor-like SNP analysis was carried out using a CFX96 Real-Time PCR Detection System (BioRad, USA) using DNA samples as described previously [21,34] with the following adjustment for barley genotyping. Each reaction with a total volume of 10 μl contained: 3 μl of template DNA adjusted to 20 ng/μl, 1 μl of the two fluorescently-labelled Universal probes mix (0.125 μM each), 1 μl of allele-specific primer mix (0.075 μM of each of two forward primers and 0.39 μM of the common reverse primer), and 2 μl of 5 × Go-Taq Master-mix (Promega, USA) with the following final concentration of components: 1.75 mM MgCl 2 , 0.2 mM of dNTP and 0.05 units of Go-Taq polymerase (Promega, USA).…”

“…In the case of wheat and barley, flowering time is generally hastened by drought (McMaster and Wilhelm, 2003;Park et al, 2016). Indeed, a recent study showed that bread-wheat TaVRN1 and TaFT1 are upregulated in leaves under drought relative to wellwatered conditions, where they are co-expressed with the CCAAT Binding Factor (CBF) transcriptional repressor TaDR1 (Zotova et al, 2019) (Fig. 2).…”

“…In other words, the TaDr1 gene can block the production and inactivate the work of proteins produced by other genes, which are no longer required by the plant or may lead to negative consequences. Depending on the state, plants regulate the activity of the TaDr1 gene, increasing or decreasing its activity [9]. Notes: 1 D -degree of dominance, %; 2 D> 100% -OD-over-dominance; 3 D=100% CDG -complete dominance of the parent's trait with a greaterexpression of the trait; 4 D= от 76 до 99% IDG -incomplete dominance of the parent with a greater expression of the trait; 5 D = от 51 до 75% PDG -partial dominance of the parent with a greater expression of the trait; 6 D=от 0 до 25% IDL -incomplete dominance of the parent with less expressed trait; 7 D = 0% CDL -complete dominance of the parent with less expressed trait; 8 D < 0% D -depression.…”
