The genetics of anorexia nervosa collaborative study: Methods and sample description

Abstract: These results provide context for the genotyping of the first 200 families by the Center for Inherited Disease Research. We will analyze our first 200 families for linkage, complete recruitment of roughly 400 families, and then perform final linkage analyses on the complete cohort. DNA, genotypes, and phenotypes will form a national eating disorder repository maintained by NIMH and available to qualified investigators.

“…The second inclusion criterion required a lifetime DSM-IV-TR diagnosis of AN with or without amenorrhea, which resulted in the exclusion of four participants. The amenorrhea criterion was waived for this analysis to be consistent with recent literature in AN 11 and is based on the replicated observation that individuals with and without amenorrhea do not differ in meaningful ways. 12,13 The final inclusion criterion required complete discharge and year 1 follow-up information.…”

“…Despite the efforts of many [16, 20, 42, 53, 54], the community’s collective understanding of AN genetic susceptibility and its influence on prognosis remains limited. The phenotypic expression of a genotype is dependent on an array of host factors including genetic background, age and gender, developmental-, physiological- and pathological- conditions, as well as environmental factors such as dietary nutrition, exercise, and nicotine.…”

“…Polyunsaturated fatty acid and eicosanoid (metabolites) assays were performed using banked plasma from convenience samples [42, 43]. Whole blood was collected in EDTA vacutainer tubes and plasma was separated by centrifugation at 1350 rpm for 10 minutes and removed using pipet.…”
