The genome sequence of the soft-rot fungusPenicillium purpurogenumreveals a high gene dosage for lignocellulolytic enzymes

Abstract: The high lignocellulolytic activity displayed by the soft-rot fungus Penicillium purpurogenum has made it a target for the study of novel lignocellulolytic enzymes. We have obtained a reference genome of 36.2 Mb of non-redundant sequence (11,057 protein-coding genes). The 49 largest scaffolds cover 90% of the assembly, and Core Eukaryotic Genes Mapping Approach (CEGMA) analysis reveals that our assembly captures almost all protein-coding genes. RNA-seq was performed and 93.1% of the reads aligned to the assemb… Show more

“…Furthermore, Camassol and Dillon (2009) reported maximum xylanase activity of 9.95 ± 1.53 U/gds of bagasse and wheat bran in combination under solid state fermentation using Penicillium echinulatum. Apart from optimizing strategies, for the enhanced production of hemicellulolytic enzymes, cloning and expression studies have also been carried out to increase the potential enzyme secretion for lignocellulose degradation to valued products (Mardones et al, 2018;Echeverria and Eyzaguirre, 2019).…”

“…Next-generation sequencing of the P. citreonigrum strain IMI92228 yielded a total of 7611 Mbp, and the final assembly was found to contain 27 Mbp in 79 scaffolds. The total length of scaffolds and estimated genome size were relatively small compared to previously reported genomes of the Penicillium species [24][25][26][27], although these scaffolds code 97.3% of single copy genes that were conserved in Eurotiomycetes, which is enough to search for the homologous gene cluster that corresponds to the CTV biosynthetic cluster of A. terreus. The estimated genome size may be underestimated because only short inset sizes were estimated, and short read analysis may be insufficient to evaluate repeat regions and transposable elements of the genome.…”

“…The composition of AP wheat straw was analyzed by sequential fractionation. In this process, the contents of cellulose, hemicellulose, and Klason lignin were determined through a quantitative acid hydrolysis [ 17 ]. In brief, the solid residue (300 mg) was treated for 1 h with 3 mL of 13.5 M sulfuric acid (H 2 SO 4 ) at 30 °C and 150 rpm.…”
