The heat shock protein 40 LeDnaJ regulates stress resistance and indole-3-acetic acid biosynthesis in Lentinula edodes

Abstract: DnaJ proteins, termed heat shock proteins based on their molecular weight, function as molecular chaperones that play critical roles in regulating organism growth and development as well as adaptation to the environment. However, little has been reported on their gene function in higher basidiomycetes. Here, the heat shock protein 40 (LeDnaJ) gene was cloned and characterized from Lentinula edodes. RNA interference was used to explore the function of LeDnaJ in response to heat stress and Trichoderma atroviride… Show more

“…The result is consistent with the expression of Hsp70 genes in other mushrooms (Chen et al 2017;Wang et al 2018a;Zhang et al 2016;Zou et al 2018). The silencing of Hsp40 gene, LeDnaJ, in L. edodes defected its resistance to heat stress (Wang et al 2018b), while the over-expression of LeDnaJ gene in L. edodes S606 conferred the strain better tolerance to heat stress (Wang et al 2018a). Overexpression of an Hsp100 gene, PsHsp100, from Pleurotus sajor-caju complemented a thermotolerance defect in hsp104 mutant S. cerevisiae (Lee et al 2006).…”

“…According to our previous studies, it was documented that the RNAi of the LeDnaJ07 decreased mycelial growth and the resistance to heat stress, while LeDnaJ07 overexpression enhanced the thermotolerance of L. edodes heatresistant strain S606 (Wang G.Z. et al, 2016, Wang et al, 2018b. However, it is worthy of study whether LeDnaJ07 overexpression regulates the resistance of other L. edodes strains to stresses or not, how LeDnaJ07 regulates the response of L. edodes to thermotolerance.…”

“…To test the effect of LeDnaJ07 overexpression on the resistance of L. edodes against heat stress, thermotolerance assays, involving short-and long-term heat stresses, were performed according to our previous method (Wang et al, 2018b). In the short-term heat stress group, the stress temperature and recovery time were 38 • C and 25 d, respectively.…”
