Summary. Separation of ovine seminal plasma proteins in the disc electrophoretic system revealed a consistent pattern of sixteen fractions with some variation among six rams in the relative amount of minor components, whereas bovine seminal plasma had twelve fractions. In both species, the major seminal plasma components had electrophoretic mobilities somewhat less than did the albumin band of the corresponding blood plasma. The prealbumin region of the seminal plasma electrophoretograms contained four fractions in the ovine samples and three fractions in the bovine samples. Storage ofdiluted ovine and bovine semen at 5\s=deg\C did not alter the total protein concentration in the seminal plasma. Standard disc electrophoretic patterns showed that two of the sixteen ovine seminal plasma protein fractions disappeared during such storage. No changes were evident in either the standard or basic electrophoretograms of bovine seminal plasma during storage of semen. Standard disc electrophoretograms of ovine and bovine sperm extracts showed many lightly staining bands, but no progressive changes were evident in these electrophoretic patterns during storage of semen.

Bombardment of bovine spermatozoa with glass beads and subsequent extraction in phosphate buffered sodium chloride yielded soluble sperm-specific proteins (approximately 10% yield) sufficient for physicochemical analysis.Moving boundary electrophoresis revealed three sperm-specific components with mobilities of 2\m=.\0, 3\m=.\8and 5\m=.\1x10-5 cm2 volt-1 sec-1 at pH 8\m=.\6in barbital (\ g=m\ = 0\m=.\10). The proportions of these components were 20, 50 and 30 %, respectively. Sedimentation velocity ultracentrifugal analysis revealed at least three sedimentation gradients. The two major gradients had S20 values of 1\m=.\7and 12\m=.\6.The third exhibited polydispersion. Diffusion coefficients of 4\m=.\2and 10\m=.\2x 10-77 cm2 sec-1 were calculated for two sperm-specific antigens by an agar-gel-diffusion method.Three protein fractions were isolated by chromatographic elution from diethylaminoethyl (deae) cellulose with an ionic gradient. The first protein fraction eluted was not bound to the deae cellulose. It migrated toward the cathode in agar-gel electrophoresis at pH 8\m=.\4, and was firmly bound to carboxymethylcellulose at pH 6\m=.\0, 7\m=.\6and 11\m=.\0. A second protein fraction from the deae cellulose appeared just after the ionic gradient commenced, and a third protein fraction appeared when the ionic gradient approached 0\m=.\2m-sodium chloride.
