The Human Antibacterial Cathelicidin, hCAP-18, Is Bound to Lipoproteins in Plasma

Abstract: Cathelicidins are a family of antibacterial and lipopolysaccharide-binding proteins. hCAP-18, the only human cathelicidin, is a major protein of the specific granules of human neutrophils. The plasma level of hCAP-18 is >20-fold higher than that of other specific granule proteins relative to their levels within circulating neutrophils. The aim of this study was to elucidate the background for this high plasma level of hCAP-18. Plasma was subjected to molecular sieve chromatography, and hCAP-18 was found in dis… Show more

“…This difference is probably due to lipoproteins present in the culture medium used for growth inhibition assays. As serum components compete with membranes for the binding of membrane-active peptides [45][46][47], they should indeed increase their apparent MICs.…”

“…Cathelin affinity-purified rabbit antiserum against recombinant hCAP18 14 was used at 1:500 dilution as earlier described 10 according to the indirect peroxidase method using a Vectastain kit (Vector Laboratories, Burlingame, CA). To ascertain the specificity of the staining, immunoabsorption was performed as earlier reported.…”

“…hCAP18 and NGAL are known to be expressed in epithelial cells as well as in neutrophils and excreted in plasma, urine, or salivary glands [39][40][41]. We measured plasma concentrations of LL-37, an active form of hCAP18, and NGAL in neonates and adults by ELISA (Fig.…”
