The human rod photoreceptor cGMP phosphodiesterase β‐subunit

Abstract: cDNA clones encoding the @bunit of the photoreceptor cGMP phosphodiesterase (PDE) were isolated from a human retina library and their sequence was determined. The encoded polypeptide consists of 854 amino acid residues with a calculated molecular mass of 98,416 Da. Alignment of the deduced amino acid sequence with the earlier analysed a-, p-and a'-subunits of bovine and mouse PDEs demonstrates a high homology. Two overlapping recombinant 1 phage clones containing 26 kb of the human PDE/%ubunit gene were isolat… Show more

“…The region near the N terminus of CyaB1 protein is similar to the N-terminal regions of the cyclic GMP (cGMP)-stimulated cGMP phosphodiesterases. This region of CyaB1, which contains 270 amino acids, shows 21.6% identity to the bovine cGMP-stimulated phosphodiesterase (44), 19.3% identity to the bovine cGMP-binding, cGMP-specific phosphodiesterase (32), and 16.7% identity to the beta subunit of the human photoreceptor cGMP phosphodiesterase (27). In mammals, this region is proposed to be an allosteric cGMP-binding region of phosphodiesterases (32).…”

“…Thus, human cone PDEcd amino acid sequence deduced from the cDNA structure consists of 858 residues with calculated molecular mass of 99169 Da. Human cone PDE~' has 89.1% and 82.7% homology to bovine [6] and chicken [13] cone PDEa', respectively; 63.8% homology to human rod PDE a subunit [14]; and 64.5% homology to human rod PDE 13 subunit [7]. Human cone PDEct', like mammalian visual PDEs, has an internal homologous repeat of the amino acid sequence in its N-terminal part.…”

“…The last 4 residues at the C-terminus conform to the CAAX consensus motif for multistep posttranslational processing covering lipidation, proteolysis and carboxymethylation [15]. Sequence analysis of genomic clones and comparison of the structure obtained with the corresponding sequences of bovine cone PDEct' cDNA [6] and human rod PDE [3-subunit gene [7] indicated the presence of fourteen N-terminal exons and 5'-flanking region. Table 1 presents data available on the exon/intron organization of this 5' part of human PDEa' gene.…”
