The Bacillus subtilis cytochrome‐c oxidase

Abstract: The structural genes of cytochrome‐c oxidase in Bacillus subtilis have been isolated and sequenced. Five genes, ctaB–F, are closely spaced. ctaC, ctaD, ctaE and ctaF are the genes for subunits II, I, III and IVB, respectively. ctaB, which may encode an assembly factor, is separated and upstream from the others. In comparison to its mitochondrial counterparts, subunit I has an extended C‐terminus with two additional transmembrane segments, whereas subunit III has lost two such segments from its N‐terminus. The … Show more

“…A sequence alignment. The sequences of subunits I and 111 of the the haem-containing copper oxidases from E. coli (CyoB and CyoC; Chepuri et al, 1990) and Bacillus subtilis (CtaC and CtaD;Saraste et al, 1991b) are aligned with SoxM. Putative transmembrane regions are hatched (note that CyoB has an extra segment labelled TMO at it N-terminus), and amino acids which are invariant in all known haem-containing copper oxidases are boxed.…”

“…A water soluble CuA domain was generated using the polymerase chain reaction (PCR), with as a template the genomic B. subtilis clone i110, carrying the eta gene cluster [12]. In this construct the ATG start codon has been introduced as a NcoI site (underlined) in the aminoterminal primer (no.…”

“…The caa 3 -type COX is expressed during growth on nonfermentable substrates such as succinate (7). It is also a typical proton-pumping hemecopper oxidase and is encoded by the ctaCDEF genes (15). Cytochrome bd can be found in B. subtilis cells grown with glucose (11,16,19) and is a QOX that does not pump protons (11) (see also Refs.…”
