The aim of this study was to obtain essential oil (LNEO) from the Laurus nobilis L. plant, and to prepare LNEO-loaded poly lactic-co-glycolic acid (PLGA) nanoparticles (NPs) as an approach in cancer treatment. The components of the obtained LNEO were analyzed using GC-MS. The LNEO-NPs were synthesized by the single-emulsion method. The LNEO-NPs were characterized using UV-Vis spectrometry, Dynamic Light Scattering (DLS), Scanning Electron Microscopy (SEM), and a DNA binding assay, which was performed via the UV-Vis titration method. According to the results, the LNEO-NPs had a 211.4 ± 4.031 nm average particle size, 0.068 ± 0.016 PdI, and −7.87 ± 1.15 mV zeta potential. The encapsulation efficiency and loading capacity were calculated as 59.25% and 25.65%, respectively, and the in vitro drug release study showed an LNEO release of 93.97 ± 3.78% over the 72 h period. Moreover, the LNEO was intercalatively bound to CT-DNA. In addition, the mechanism of action of LNEO on a dual PI3K/mTOR inhibitor was predicted, and its antiproliferative activity and mechanism were determined using molecular docking analysis. It was concluded that LNEO-loaded PLGA NPs may be used for cancer treatment as a novel phytotherapeutic agent-based controlled-release system.

Background: The present study aimed to identify an NRLs (necroptosis-related long noncoding RNAs [lncRNAs]) signature for the prognosis of colon adenocarcinoma (COAD) and to investigate its potential relationship with clinical characteristics and immune microenvironment. Methods: Clinical information of patients with COAD and RNA sequence data were downloaded from The Cancer Genome Atlas (TCGA) database. Necroptosis-related genes (NRGs) were obtained from the GeneCards website. Co-expression analysis was performed to identify NRLs. By using the “limma” package in R software, differentially expressed NRLs (DE-NRLs) were screened from the identified NRLs. A prognostic NRLs signature was established based on the results of univariate Cox regression analysis, LASSO algorithm, and multivariate Cox analysis. Survival analysis and area under the curve (AUC) of receiver operating characteristic (ROC) were used to evaluate the prognostic efficacy of this signature. Clinical correlation and independent prognostic factors were then assessed, and a predictive nomogram was then constructed. Finally, gene set enrichment analysis (GSEA) was conducted to determine the potential molecular mechanisms. Immune analysis was performed to analyze the tumor microenvironment and to predict the immune response of patients with COAD. Lastly, experiments were conducted to validate the expression and function of NRLs involved in the prognostic signature. Results: Three NRLs were identified, and a prognostic signature was constructed from them. According to the risk score calculated using this signature, patients were classified into high-risk and low-risk groups. The signature exhibited a strong ability to predict survival, and the risk score was confirmed as an independent prognostic factor. GSEA showed differences in specific molecular pathways involved in immune response, mitochondrial function, and energy metabolism. The two groups also showed significant differences in immune cell infiltration, immune function, and immune checkpoint gene expression. The high-risk group showed higher immunogenicity and may benefit from immunotherapy. Lastly, in vivo experiments validated the expression and function of NRLs involved in the prognostic signature. Conclusions: The prognostic signature of NRLs developed in this study can facilitate the clinical diagnosis and treatment of patients with COAD and may serve as a valuable reference for in-depth studies of functional mechanisms of NRLs in the immune microenvironment.

Background: Anti-cancer agents encapsulated in nanoparticles (NPs) can result in higher efficiency. Kombucha is a fermented tea beverage, and previous reports support its anti-cancer properties. Objectives: The present study aimed to evaluate the anti-cancer and anti-inflammatory properties of poly (lactic-co-glycolic acid) loaded Kombucha NPs (PLGA-K-NPs) against the A2780 human ovarian cancer cell line. Methods: The antioxidant activity was analyzed using ferric reducing ability of plasma and 2,2’-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) assays, along with the measurement of catalase (CAT) gene expression. The gene expression of three interleukins (IL-1β, IL-6, and IL-10) was also determined to demonstrate the anti-inflammatory properties of NPs. Results: The results revealed the antioxidant effects of PLGA-K-NPs on the studied cell lines by increasing Fe3+ reduction, inhibiting the free radical formation (P<0.001), and increasing the expression of the CAT gene (P<0.001). In addition, NPs could significantly elevate the gene expression of IL-10 (P<0.01) as an anti-inflammatory cytokine at a 40 µg/mL concentration, while reducing the expression of IL-1β and IL-6, and inflammatory cytokines at all tested concentrations (P<0.01). Conclusion: According to the obtained results, PLGA-K-NPs have anti-inflammatory and anti-oxidant properties, therefore, they can be considered as a compound in the treatment of ovarian cancer. However, it needs to be further investigated in animal studies to clarify more details.
