The Drosophila Toll Signaling Pathway

Abstract: The identification of the Drosophila melanogaster Toll pathway cascade and the subsequent characterization of TLRs have reshaped our understanding of the immune system. Ever since, Drosophila NF-κB signaling has been actively studied. In flies, the Toll receptors are essential for embryonic development and immunity. In total, nine Toll receptors are encoded in the Drosophila genome, including the Toll pathway receptor Toll. The induction of the Toll pathway by Gram-positive bacteria or fungi leads to the activ… Show more

“…Four of five known profile χ immune genes are Toll-like proteins and peptidoglycan recognition proteins. Toll-pathway and peptidoglycan recognition proteins are used for defense against bacteria and fungi (25,26), and therefore the profile χ expression pattern and associated genomic blocks might be related to immunity against these microbiota. Nonconserved genomic regions were underrepresented significantly in all three profiles (P = 7 × 10 −5 ).…”

“…4). The mussel genes expressed in response to bacteria likely constitute a complete pathway from TLR to NF-jB, similar to Toll/TLR pathways traced in model species; however, the involvement and the role of intermediate transcript elements is still hypothetical as based on functional analogy (Valanne et al, 2011). The expression of MgIKK-1 (IKK-a/b sub-group) and MgNEMO (IKK-c) genes are both induced by injection of bacteria, however with less intensity for MgNEMO.…”

“…The cell signaling pathway triggered by Gram-positive bacteria and fungal infections, known as Toll pathway, has been described completely in insects (Valanne et al, 2011). Another cell signaling cascade, the IMD (immune deficiency) pathway regulates the insect response to Gram-negative bacterial infections (Lemaitre and Hoffmann, 2007).…”

“…For example, in D. melangastor, the Toll-like receptor pathway is involved in the response to microbial infection. On immune stimulation, protease cascades lead to the activation of the cytokine Spatzle (33). In humans, the related pathway involves the Toll-like receptor (IL1-R1) and its respective ligand (IL-1) (34).…”
