TheLegionella pneumophilametaeffector Lpg2505 (SusF) regulates SidI-mediated translation inhibition and GDP-dependent glycosyltransferase activity

Abstract: 19Legionella pneumophila, the etiological agent of Legionnaires Disease, employs an arsenal of 20 hundreds of Dot/Icm-translocated effector proteins to facilitate replication within eukaryotic 21 phagocytes. Several effectors, called metaeffectors, function regulate the activity of other 22 Dot/Icm-translocated effectors during infection. The metaeffector Lpg2505 is essential for L. 23 pneumophila intracellular replication only when its cognate effector, SidI, is present. SidI is a 24 cytotoxic effector that i… Show more

“…1 A ). DT and exoA are ADP-ribosylating enzymes that inactivate EEF2 at a conserved diphthamide residue, whereas sidI acts as a transfer RNA mimic that inactivates EEF1 and ribosome components ( Subramanian et al, 2022 Preprint ; Joseph et al, 2019 Preprint ). A small molecule inhibitor of EEF1, didemnin B (DDB, also known as Aplidine; Crews et al, 1994 ; Fig.…”
