TheO-GlcNAc transferase OGT is a conserved and essential regulator of the cellular and organismal response to hypertonic stress

Abstract: The conserved O-GlcNAc transferase OGT O-GlcNAcylates serine and threonine residues of intracellular proteins to regulate their function. OGT is required for viability in mammalian cells, but its specific roles in cellular physiology are poorly understood. Here we describe a conserved requirement for OGT in an essential aspect of cell physiology: the hypertonic stress response. Through a forward genetic screen in Caenorhabditis elegans, we discovered OGT is acutely required for osmoprotective protein expressio… Show more

“…1C) that recruit OGT to specific substrates (3,(30)(31)(32)(33)(34). Recent evidence suggests OGT can regulate Caenorhabditis elegans physiology independently of its catalytic functions (35,36); however, given that OGT is not essential in C. elegans (37), it remains unclear whether OGT's binding interactions are essential as well as whether they broadly regulate mammalian physiology independent of OGT's enzymatic activity (38,39).…”

“…However, evidence from thousands of RNAseq datasets does not reveal evidence for APA of gpdh-1 itself (S teber et al . 2019) and genetic studies show that gpdh-1 null alleles can still activate hypertonic adaptation (U rso et al . 2020).…”

“…In addition to catalyzing both glycosylation and cleavage reactions, OGT interacts with binding partners ( Figure 1C) that recruit OGT to specific substrates (3,(30)(31)(32)(33)(34). Recent evidence suggests OGT may regulate C. elegans physiology independent of its catalytic function (36,37); however, given that OGT is not essential in C.…”
