The Urtica dioica Agglutinin Is a Complex Mixture of Isolectins

Abstract: Rhizomes of stinging nettle (Urtica dioica) contain a complex mixture of isolectins. Ion exchange chromatography with a high resolution fast protein liquid chromatography system revealed six isoforms which exhibit identical agglutination properties and carbohydrate-binding specificity and in addition have the same molecular structure and virtually identical biochemical properties. However, since the U. dioica agglutinin isolectins differ definitely with respect to their amino acid composition, it is likely tha… Show more

“…Test Compounds-The mannose-specific plant lectins from Hippeastrum hybrid (HHA) and from G. nivalis (GNA) and the N-acetylglucosamine-specific plant lectin from U. dioica (UDA) were derived and purified from these plants as described previously (15,16) and kindly provided by Prof. E. J. M. Van Damme and Dr. W. Peumans (Ghent, Belgium). Pradimicin A (PRM-A) was obtained from Prof. T. Oki and Prof. Y. Igarashi (Toyama, Japan).…”

“…The chitin-binding domain consists of one Ser and two aromatic amino acid residues located at the intercysteine loop 3 and one aromatic residue at loop 4 ( Figure 1 ). This conserved chitin-binding domain can also be found in class I chitinases, such as Cht-2 from rice Oryza sativa (Takakura et al, 2000) and urtica dioica agglutinin (UDA) from the stinging nettle Urtica dioica (Van Damme et al, 1988). …”

“…PRM-A (molecular weight, 838) was isolated and purified from the fermentation broth of the Actinomyces species Actinomadura hibisca TP-A0016 (12,26). The mannose-specific plant lectins GNA, HHA, and UDA were derived and purified from these plants as previously described (33,34). Polyvinyl alcohol sulfate (PVAS) was kindly provided by S. Görög (Budapest, Hungary).…”
