The IL-6R α chain controls lung CD4+CD25+ Treg development and function during allergic airway inflammation in vivo

Abstract: The cytokine IL-6 acts via a specific receptor complex that consists of the membrane-bound IL-6 receptor (mIL-6R) or the soluble IL-6 receptor (sIL-6R) and glycoprotein 130 (gp130). In this study, we investigated the role of IL-6R components in asthma. We observed increased levels of sIL-6R in the airways of patients with allergic asthma as compared to those in controls.

“…Since IL-6 production has been linked to T reg cell downregulation [17,18], we treated mice with anti-IL-6R antibody before and after vaccination to block IL-6 production in vivo. Neutralization of soluble IL-6 did not have any effect on vaccination outcome or T reg cell numbers (not shown).…”

“…This phenomenon is regulated by complex DC-CD4 + T cell interactions [34]. Recently, Doganci et al [17] studied cytokine production and T reg cell function in a model of OVA-induced allergy airway inflammation, finding that neutralization of membranebound IL-6R reversed T reg cell suppression. In this report, we have confirmed, in a model of infectious disease and vaccination, that IL-6 is responsible for the down-regulation of T reg cell numbers.…”

“…2C (lower panel). Lung CD4 + cells (10 5 /well) were then labeled with CFSE (Molecular Probes, Eugene, OR) in the presence of soluble anti-CD3 (2.5 lg/mL) and anti-CD28 antibodies (1 lg/mL) overnight and as previously described [42]. After 24 h, the starting incorporated fluorescence was recorded (M1).…”
