The impact of BNT162b2 mRNA vaccine on adaptive and innate immune responses

Abstract: The mRNA-based BNT162b2 vaccine from Pfizer/BioNTech was the first registered COVID-19 vaccine and has been shown to be up to 95% effective in preventing SARS-CoV-2 infections. Little is known about the broad effects of the new class of mRNA vaccines, especially whether they have combined effects on innate and adaptive immune responses. Here we confirmed that BNT162b2 vaccination of healthy individuals induced effective humoral and cellular immunity against several SARS-CoV-2 variants. Interestingly, however, … Show more

“…Local, such as so-called Covid arm [ 47 ], and distal inflammatory reactions in the heart and brain have been reported with the mRNA-LNP platform [ 48 53 ]. The robust inflammation driven by the ionizable lipid component of the LNPs [ 7 ] likely contributed to the innate and adaptive immune reprogramming recently reported with this vaccine platform [ 54 ]. Thus, we think that more basic and translational research would be needed before this platform gets full approval for human use.…”

“…Immune receptor TLR3,7,8 and RIG-1 are intimately involved in targeting such secondary structures and their response is RNA modification dependent 19 . This has been described as "reprogramming the innate immune and adaptive immune response" 20 . This may explain the increased rate of Herpes Zoster, HHV6/HHV7 (Pityriasis rosea) and Epstein-Barr reactivation post mRNA vaccination [21][22][23][24][25][26] .…”

“…There is limited literature describing fidelity of mRNA vaccine derived spike protein expression. The best in-vivo evidence of heterogeneous translation of vaccine derived spike protein is from Figure 2C While these truncated or elongated promiscuous translation products may be rare events, this does bring to focus the question of dosage and spike protein duration with mRNA vaccines that ablate many components of the innate immune system 20 .…”
