The Impact of p53 on Aristolochic Acid I-Induced Gene Expression In Vivo

Abstract: Exposure to aristolochic acid (AA) is linked to kidney disease and urothelial cancer in humans. The major carcinogenic component of the AA plant extract is aristolochic acid I (AAI). The tumour suppressor p53 is frequently mutated in AA-induced tumours. We previously showed that p53 protects from AAI-induced renal proximal tubular injury, but the underlying mechanism(s) involved remain to be further explored. In the present study, we investigated the impact of p53 on AAI-induced gene expression by treating Trp… Show more

“…In our hands, TNF from the proximal nephron augments the proportions of TECs stuck in the G2/M phase with consequent exacerbations in kidney fibrosis after chronic AAN. p53 is a tumor suppressor that regulates cell death and metabolism in response to injurious stimuli, including hypoxia and DNA damaging agents, 40 and plays a fundamental role in instigating proximal tubular cell injury. 41 In turn, post-translational activation of p53 promotes the apoptosis of TECs after AA injection.…”

“…Some empirical data and cytotoxicity tests of aristolochia species show potential directions as anti-cancer. Thus, preliminary research efforts on the cytotoxic effects of various aristolochia species can be an initial study in the discovery and development of anti-cancer substances from natural ingredients (16) .…”
